Cytotoxicity Assay

(Antibody-dependent Complement-mediated)

Various methods are used for the analysis of antibody-mediated, complement-dependent cytotoxicity. Xeno utilizes flow cytometric analysis of target cells to establish the serum concentration which kills 50% (IC50) of the target cells. The assay utilizes target cells from the donor species incubated with heat-inactivated and serially-diluted serum from the host. The serum is subsequently activated by addition of exogenous complement. The IC50 is determined by the change in labeling pattern of cells incubated with fluorescein diacetate (FDA) for live cells and propidium iodide (PI) for dead cells. The method is useful in assessing changes in antibody titer associated with tissue or organ transplants. For information regarding our other contract research services, visit our capabilities page.
